if you were doing audio you could use the simple superposition (phase cancelling) aspects of kareoke vocal removal.
with this melody-less version you would then invert its phase and sum that with a mono version of the original song, thus leaving you with just the melody, which you can then use frequency analysis to determine the fundamentals of the melody and notate it.
