Melody, in general, is the tune, as opposed to harmony, which is the chords. However, when people sing "harmony," or harmonize with the melody, what they're doing is singing another "melodic" line, or melody, that creates "harmony" with the original tune. That poses a problem for you. If you can get a song that has just one person singing a tune, then you can define the melody as the bit that the singer sings. If more than one person is singing, and each singer is singing a different line, then you'll have a real challenge determining which line is the official "melody." Your ear will tell you, as may the volume of each voice, and other parts of the song may also make it obvious which is the main melody, but instructing a computer to hear one line as melody and the other as not melody could be tricky.
