There are colonial acts dating back to the early part of the 19th century which make reference to "The King's Highway" in Upper and Lower Canada. Like other references to the monarch the designation is specific to the gender of the sovereign on the throne at any time. Thus in 1952 all KCs became QCs, the King's Honorary Physician became the Queen's Honorary Physician etc. There are a couple of good websites on line that chronicle the history of Canadian road signs.
